To observe the response of the spleen in albino rat to Paragonimus iloktsuenensis infection with the distribution of the mast cells, the experimental animals were given a number of 20 metacercariae and have fed for 3 days, 3 weeks and 4 weeks respectively. Immediately after the end of the experimental period, the splenic tissues were fixed in 10% neutral formalin and embedded in paraffin. Their sections were stained by hematoxylin and eosin for histological observations, and by toluidine blue for the demonstration of the mast cells. All of the sections were examined under the light microscope and the results obtained were as follows: 1. In the early stages of the infection, the lungflukes were found in the abdominal cavity and then from the thoracic cavity and the lung parenchyma individually. 2. No noticeable changes in the spleen were recognized on the staining of hematoxylin and eosin. 3. There were no mast cells in the splenic parenchyma as well as in the connective tissue of splenic capsule and trabeculae. 4. Differentiation process of the mast cells and their relationship with anaphylactic response were discussed on the basis of the above findings.
